Research Interests:  Development of strategies for the immunotherapy of relapsed acute lymphoblastic leukaemia (ALL).  Our studies have focused on the growth of dendritic cells (DC) from patients with ALL at presentation or relapse.  We have shown that functional dendritic cells may be obtained from children with ALL and used to stimulate the proliferation of allogeneic T-cells.  Initially we studied the mixed lymphocyte reaction and the ability of T-cells to proliferate in response to presentation of CMV peptides by both normal and leukaemic dendritic cells.  In initial experiments we showed that leukaemic DC could induce proliferation of T-cells with anti-leukaemic activity.  Following Research and Ethics Committee approval to obtain more extensive cell samples of from children with ALL as well as their related and unrelated donors, we are aiming to further characterise the type of dendritic cells produced in these children, their functional characteristics and the nature of the T-cell response to them.  If we are able to show that in a further cohort of number of children that we can reliably produce anti-leukaemic responses then we will apply to the Research and Ethics Committee for permission to undertake clinical studies.  The approaches used in this study have much in common with those used to educate T-cells to eliminate viral infections in patients post-transplant.
